Detailed illumination system
The illumination system basically consists of a power supply, LEDs, and filters. For regular voltage and calcium optical mapping, there should be 3 red LEDs and 3 green LEDs on magnetic bases. Each LED should also have a collimating lens and a filter attached. The filters should be 640nm and 550nm for red and green, respectively. There are 4 outputs on the face of the power supply, the top pair (red and black) is one channel and all 3 LEDs of red or green should be connected in series to that channel. The opposite color should be attached to the other channel, the pair of outputs below. The power switch for the power supply is on the back side near the power cord. There are 3 knobs on the face, the top two control the current to the channels, the lower one is used to select things on the menu. Push the knob in to move the menu selector down, turn left or right to change the selected item. There are 4 pages that can be selected by turning the knob when the selector is at the top of the page. The first page is for setup, ignore it. the second page uses internal timing to switch between the channels. The third page uses external timing to switch between channels. This is the setting we generally use for experiments as it coordinates the timing of the camera with the illumination system. To use this setting, ensure that the cable from camera frame readout is attached to the input at the front of the power supply. As a note, the camera and the program must both be running for the lights to be on with this setting. The fourth and final page is used for testing the channels. You can turn each channel on independently to adjust LED positions for optimal illumination of your point of interest.
